Biological Background: KIF4A Function and Localization
- KIF4A, also known as Chromokinesin-A, is a chromosome-associated kinesin motor protein encoded by the KIF4A gene in humans.
- It acts as a microtubule-based motor protein that translocates PRC1 to the plus ends of interdigitating spindle microtubules during mitosis, essential for central spindle midzone and midbody formation and successful cytokinesis. Related references: PMID:15297875 PMID:15625105
- KIF4A is an iron-sulfur (Fe-S) cluster binding protein that plays a role in chromosome segregation during mitosis. Related references: PMID:29848660
- Subcellular localization includes the nucleus matrix, cytoplasm, cytoskeleton, spindle, midbody, and chromosomes, reflecting its dynamic distribution during the cell cycle.
- Tissue specificity: highly expressed in hematopoietic tissues (fetal liver, spleen, thymus, adult thymus, and bone marrow); lower levels in heart, testis, kidney, colon, and lung.
- The protein contains coiled-coil domains and ATP-binding motifs, and is subject to phosphorylation and ubiquitination (Ubl conjugation).
- Mutations in KIF4A are associated with X-linked intellectual disability (MRX100/XLID100), highlighting its role in neurodevelopment.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an N-terminal peptide (aa 1–100) of human KIF4A; verify sequence conservation in mouse and rat to support observed cross-reactivity.
- For Western blot, the predicted molecular weight is ~140 kDa; use an appropriate gel percentage (e.g., 8 % SDS‑PAGE) and protein markers for high-molecular-weight targets.
- For IHC-P, antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 9.0) should be optimized; validate positive staining in tissues with high KIF4A expression, such as thymus or bone marrow.
- In ELISA, consider using the immunizing peptide or recombinant KIF4A protein as a coating antigen and titrate the antibody for optimal signal-to-noise ratio.
