Biological Background: KIF23 Function and Localization
- KIF23, also known as kinesin-like protein 5 (KNSL5) or mitotic kinesin-like protein 1 (MKLP1), is a plus-end-directed motor enzyme that moves antiparallel microtubules in vitro.
- It is a component of the centralspindlin complex, essential for cytokinesis by mediating Rho-dependent signaling required for myosin contractile ring formation during cell division.
- KIF23 is required for the localization of ECT2 to the central spindle, further facilitating cytokinesis.
- The protein localizes to the nucleus, cytoplasm, cytoskeleton (spindle), and midbody, with specific localization to the midbody ring during cytokinesis.
- It is widely expressed in human tissues.
- KIF23 undergoes alternative splicing and is subject to post-translational modifications, including phosphorylation and ubiquitination.
- Mutations in KIF23 are associated with congenital dyserythropoietic anemia and hereditary hemolytic anemia.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 801-960) of human KIF23, which may provide specific recognition of the full-length protein. Consider sequence alignment to assess cross-reactivity with other species or family members.
- For Western blotting, a band at approximately 110 kDa is expected. Use appropriate positive controls such as HeLa or HEK293 cell lysates, and verify the detected band against the predicted molecular weight.
- For immunoprecipitation, cross-reactivity with human and rat samples is anticipated. Optimize antibody concentration and incubation conditions for your specific experimental system.
- For ELISA, direct coating of recombinant KIF23 protein or peptide is recommended. Include appropriate negative controls to assess background binding.
