Biological Background: KIF20B Function and Localization

  • Kinesin-like protein KIF20B, also known as M-phase phosphoprotein 1 (MPP1), cancer/testis antigen 90 (CT90), and kinesin family member 20B, is encoded by the KIF20B gene (aliases KRMP1, MPHOSPH1).
  • Functions as a plus-end-directed microtubule motor essential for completion of cytokinesis, including proper midbody organization and abscission in polarized cells. Related references: PMID:11470801, PMID:12740395
  • In neurons, KIF20B regulates polarization by mediating transport of SHTN1 and accumulation of PIP3 in growth cones, contributing to radial migration of cortical neurons and cerebral cortex growth.
  • Acts as an oncogene in bladder cancer, promoting cell proliferation, inhibiting apoptosis, and driving carcinogenic progression. Related references: PMID:17409436
  • Subcellularly localizes to the nucleus (nucleolus, nucleoplasm), cytoplasm, microtubule organizing center (centrosome), mitotic spindle and spindle pole, midbody, and neuronal projections including axons and growth cones.
  • Detected at the protein level in brain, ovary, kidney, and testis; overexpressed in bladder cancer cells. Related references: PMID:12740395, PMID:17409436
  • Belongs to the kinesin motor protein family, harboring ATP-binding and microtubule-binding domains; classified as a phosphoprotein, oncogene, and reference proteome member.

Experimental Guidance and Technical Tips

  • The immunogen design targets the C-terminal region (aa 1200-1500) of human KIF20B; consider this antibody for Western blotting (predicted band at ~211 kDa) and ELISA as validated.
  • For applications beyond WB/ELISA (e.g., immunofluorescence, immunohistochemistry), validate in the relevant sample system based on known subcellular distribution.
  • Optimize antibody dilution and blocking conditions for each assay, paying special attention to transfer efficiency for high-molecular-weight targets in Western blotting.
