Biological Background: Ki67 Function and Localization
- MKI67 (Ki-67) is a well-established proliferation marker expressed in the nucleus of actively dividing cells, with expression levels closely correlated with the cell cycle.
- It associates with mitotic chromosomes during early mitosis, forming a characteristic brush-like structure that acts as a steric and electrostatic barrier to prevent chromosome clustering. Related references: PMID:27362226
- The high net charge of Ki67 contributes to its surfactant-like properties, dispersing chromosomes and facilitating independent motility. Related references: PMID:27362226
- In anaphase, Ki67 switches to a chromosome attractant, promoting chromosome clustering through dephosphorylation and RNA-dependent liquid-liquid phase separation.
- It binds DNA, particularly supercoiled and AT-rich sequences, and interacts with premature ribosomal RNAs to modulate chromosomal organization.
- Ki67 localizes to the chromosome surface, nucleolus, and nucleus. Post-translational modifications include acetylation and phosphorylation.
- The protein has a predicted molecular weight of ~350 kDa, with alternative splicing leading to variants of 359 kDa.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 2271–2370; consider including both positive and negative KO cell lysates for validation in WB.
- For WB, use a low-percentage gel (e.g., 6–8%) and extended transfer times due to the large protein size (~350 kDa).
- In IHC-P, nuclear Ki67 staining requires effective antigen retrieval; citrate buffer (pH 6.0) with heat is commonly used.
- For IF/ICC, a permeabilization step with 0.1–0.5% Triton X-100 is recommended to access nuclear antigens.
- This antibody is validated for knockout studies; verify specificity in your model system by comparing wild-type and KO samples.
