Biological Background: Ki67 Function and Localization
- Proliferation marker protein Ki-67 (MKI67) associates with the surface of mitotic chromosomes and acts both as a chromosome repellent during early mitosis and a chromosome attractant during late mitosis. Related references: PMID:27362226, PMID:32879492, PMID:35513709
- During early mitosis, Ki-67 relocalizes from nucleoli to the chromosome surface and forms extended brush-like structures that prevent chromosome collapse by providing a steric and electrostatic charge barrier. Related references: PMID:27362226
- At mitotic anaphase, dephosphorylation of Ki-67 and exposure of a conserved basic patch induce RNA-dependent liquid-liquid phase separation on the chromosome surface, promoting chromosome clustering and exclusion of large cytoplasmic particles from the future nuclear space. - For IF/ICC, consider optimizing cell fixation and permeabilization to preserve nucleolar and mitotic chromosome staining. Proliferating cells such as HeLa or Jurkat can serve as positive controls.
- For ELISA, validate antibody reactivity using recombinant Ki67 fragments or whole-cell lysates from proliferating versus quiescent cells.
- Since the antibody recognizes human Ki67, cross-reactivity with other species is not expected unless there is high sequence homology in the immunogen region.
