Biological Background: Ki67 Function and Localization

  • Ki67, encoded by the MKI67 gene, is a well-established proliferation marker protein (also known as Antigen KI-67 or Antigen Ki67).
  • During early mitosis, Ki67 relocalizes from nucleoli to the chromosome surface, forming extended brush-like structures. These highly charged brushes act as a surfactant, preventing chromosome aggregation via steric and electrostatic repulsion. Related references: PMID:27362226
  • In anaphase, Ki67 switches to a chromosome attractant, undergoing dephosphorylation and RNA-dependent liquid-liquid phase separation to promote chromosome clustering. Related references: PMID:32879492, PMID:39153474, PMID:28935370
  • Ki67 binds DNA with a preference for supercoiled and AT-rich sequences, and also interacts with premature ribosomal RNAs during anaphase. Related references: PMID:10878551, PMID:28935370
  • It may also contribute to chromatin organization, though the direct mechanism remains unclear. Related references: PMID:24867636
  • Ki67 localizes to the nucleolus during interphase and to the chromosome surface during mitosis.
  • The protein is subject to various post-translational modifications including phosphorylation, acetylation, isopeptide bond formation, and ubiquitin conjugation.

Experimental Guidance and Technical Tips

  • The recombinant immunogen covers amino acids 460-680 of human Ki67 (NP_002408.3), which may influence epitope specificity. Consider this when designing detection assays.
  • For western blot, note the high molecular weight (~359 kDa); use low-percentage SDS-PAGE gels and extended transfer times or wet transfer methods to ensure efficient transfer.
  • For IHC-P, validate antigen retrieval conditions and primary antibody dilution in the relevant sample system. Include appropriate positive (e.g., proliferating tissues) and negative controls.
  • This antibody shows cross-reactivity with mouse, enabling its use in xenograft or murine model studies; however, verify species-specific performance experimentally.
  • ELISA applications require optimization of coating and detection protocols.
