Biological Background: KHDRBS2 Function and Localization
- KHDRBS2, also known as SLM-1 or Sam68-like mammalian protein 1, is an RNA-binding protein that belongs to the KH domain-containing family and plays a critical role in alternative splicing regulation, influencing mRNA splice site selection and exon inclusion.
- It binds both poly(A) and poly(U) homopolymers, and its RNA-binding ability is inhibited upon phosphorylation by PTK6. Additionally, phosphorylation by FYN inhibits its ability to regulate splice site selection.
- KHDRBS2 induces concentration-dependent incorporation of exon in CD44 pre-mRNA via direct binding to purine-rich exonic enhancers, and regulates alternative splicing of NRXN1 in the laminin G-like domain 6 containing the conserved neurexin alternative spliced segment 4 (AS4) that is involved in synaptic targeting.
- It acts synergistically with SAM68 in NRXN1 exon skipping at AS4, but antagonistically in NRXN3 exon skipping, demonstrating context-dependent regulatory interactions within the SAM68 family.
- Subcellularly, KHDRBS2 localizes primarily to the nucleus, consistent with its role in pre-mRNA processing and transcription regulation.
- Tissue expression is highest in brain, lung, kidney, and small intestine, with weaker expression in placenta, liver, spleen, thymus, ovary, and colon, suggesting tissue-specific splicing functions.
- The protein undergoes post-translational modifications including phosphorylation and methylation, and contains SH3-binding motifs, indicating its potential role as an adapter protein for Src kinases during mitosis.
- As per UniProt keywords, KHDRBS2 is involved in mRNA processing, transcription regulation, and is classified as a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning residues 200 to the C-terminus of human KHDRBS2 (NP_689901.2), which is expected to generate antibodies recognizing the target protein in its native and denatured forms; consider verifying isoform specificity by sequence alignment.
- For Western blot, the predicted molecular weight is approximately 39 kDa; validate the observed band size in endogenous samples from highly expressing tissues such as brain or lung lysates, and include appropriate controls to confirm specificity.
- In immunohistochemistry (IHC-P), antigen retrieval conditions and antibody dilution may require optimization; consider testing a range of dilutions and including isotype controls, especially in tissues with known high expression.
- For ELISA, the polyclonal nature of the antibody may provide robust detection; however, pair compatibility with capture antibodies should be assessed in sandwich format, and cross-reactivity with mouse samples can be exploited for translational studies.
