Biological Background: KEAP1 Function and Localization
- KEAP1 (Kelch-like ECH-associated protein 1), also referred to as INrf2 or KLHL19, is a 70 kDa cytoplasmic and nuclear protein that functions as a substrate-specific adapter for the BCR (BTB-CUL3-RBX1) E3 ubiquitin ligase complex.

- As a key oxidative stress sensor, KEAP1 contains highly reactive cysteine residues that undergo non-enzymatic covalent modification by electrophiles, leading to inactivation of the ligase complex and subsequent nuclear accumulation of NRF2. - The immunogen comprises amino acids 320–510 of human KEAP1; this region may allow recognition of full-length KEAP1 and potentially conserved epitopes across species. Verify specificity in your sample system.
- For Western blotting, expect a band corresponding to the predicted molecular weight of approximately 70 kDa. Include appropriate positive and negative controls.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), KEAP1 can be detected in both cytoplasmic and nuclear compartments. Consider optimizing fixation and permeabilization steps.
- The reported cross-reactivity includes human, mouse, and rat; however, performance in each species should be validated with relevant tissues or cell lines.
