Biological Background: KEAP1 Function and Localization
- KEAP1 (Kelch-like ECH-associated protein 1), also known as INrf2 or KLHL19, functions as a substrate-specific adapter for the BCR (BTB-CUL3-RBX1) E3 ubiquitin ligase complex. Related references: PMID:14585973 PMID:15379550 PMID:15572695
- It serves as a critical redox sensor by targeting the transcription factor NFE2L2/NRF2 for ubiquitination and proteasomal degradation under basal conditions, thus maintaining low levels of cytoprotective gene expression. Related references: PMID:15601839 PMID:16006525
- Upon oxidative or electrophilic stress, reactive cysteine residues in KEAP1 are covalently modified, inactivating the ligase complex and allowing NRF2 to accumulate in the nucleus and drive phase II detoxifying enzyme expression. Related references: PMID:16006525 PMID:17127771 PMID:18251510
- Selective autophagy pathways involving SQSTM1/p62 can sequester KEAP1, leading to NRF2 activation; the BCR(KEAP1) complex also ubiquitinates SQSTM1/p62, enhancing its own degradation. Related references: PMID:20452972 PMID:28380357
- Additional substrates of the BCR(KEAP1) complex include BPTF and PGAM5, broadening its role in cellular regulation. Related references: PMID:15379550 PMID:17046835
- KEAP1 is predominantly cytoplasmic but can also localize to the nucleus, consistent with its regulatory functions.
- It is broadly expressed in human tissues, with the highest levels observed in skeletal muscle.
- Post-translational modifications include S-nitrosylation and ubiquitination, and it contains Kelch repeat domains critical for substrate recognition.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of KEAP1 is approximately 70 kDa; use appropriate positive controls from human, mouse, or rat lysates to confirm specificity.
- In immunofluorescence/ICC, consider optimizing fixation and permeabilization conditions to detect both cytoplasmic and nuclear pools of KEAP1.
- For sandwich ELISA applications, pairing with a validated capture antibody is recommended; validate the antibody pair in the relevant sample matrix.
- As a polyclonal antibody, lot-to-lot consistency may vary; it is advisable to test new lots in a pilot experiment before large-scale studies.
