Biological Background: KDM8 Function and Localization
- Bifunctional enzyme: possesses endopeptidase and 2-oxoglutarate-dependent monooxygenase activities. Endopeptidase cleaves histone N-terminal tails at methylated arginine or lysine, generating tailless nucleosomes. Preferentially targets monomethylated and dimethylated arginine residues of histones H2, H3, and H4. Upon DNA damage, cleaves H3 at monomethylated Lys-9, with H3F3A as the major target (PubMed:28847961, PubMed:28982940, PubMed:29459673). Related references: PMID:28847961, PMID:28982940, PMID:29459673
- Monooxygenase activity catalyzes (R)-stereospecific hydroxylation at C-3 of Arg-137 of RPS6 and Arg-141 of RCCD1 (PubMed:29563586). Related references: PMID:29563586
- Regulates mitosis: transcriptionally represses satellite repeats possibly by controlling H3K36 methylation at centromeric regions together with RCCD1; involved in proper mitotic spindle organization and chromosome segregation (PubMed:24981860). Related references: PMID:24981860
- Negatively regulates the cell cycle repressor CDKN1A/p21, controlling G1/S phase transition (PubMed:24740926). Related references: PMID:24740926
- Required for G2/M phase progression and regulates CCNA1/cyclin-A1 expression, promoting cancer cell proliferation (PubMed:20457893). Related references: PMID:20457893
- Modulates alpha-tubulin acetylation and cytoskeletal microtubule stability, contributing to epithelial-mesenchymal transition (PubMed:28455245). Related references: PMID:28455245
- Regulates circadian gene expression in the liver; represses CLOCK-BMAL1 transcriptional activity in a catalysis-independent manner; negatively regulates CRY1 protein stability, required for AMPK-FBXL3-induced CRY1 degradation (PubMed:30500822). Related references: PMID:30500822
- Subcellular localization: nucleus; associates with chromosomes.
- Tissue specificity: weakly expressed in most cells, but highly expressed in breast cancer cells and embryonic stem cells.
- Annotated with keywords: Alternative splicing, Chromatin regulator, Cell cycle, Biological rhythms, Iron, Metal-binding, Nucleus.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 147–416 of human KDM8 (NP_079049.2), covering a conserved region that includes part of the catalytic domain, which likely underpins cross-reactivity with mouse.
- For Western blotting, a predominant band near 47 kDa is expected; however, alternative splicing or post-translational modifications may produce additional bands, so validate in your specific sample system.
- This polyclonal antibody is suitable for ELISA; consider using it for detection of native or recombinant KDM8 in cell lysates or purified preparations.
- Lot-to-lot variability is inherent to polyclonal antibodies; it is advisable to titrate and validate each lot for your particular assay conditions.
- For functional assays, note that antibody binding may not directly inhibit KDM8’s enzymatic activities unless specifically validated.
