  • It specifically demethylates dimethylated lysine-9, lysine-27, and lysine-36 on histone H3 (H3K9me2, H3K27me2, H3K36me2) and monomethylated lysine-20 on histone H4 (H4K20me1), playing a central role in the histone code.
  • The demethylase activity of KDM7A is regulated by H3K4me3 binding: in the presence of H3K4me3, it demethylates H3K27me2 but not H3K9me2; in the absence of H3K4me3, it demethylates H3K9me2.
