Biological Background: KDM5A Function and Localization
- Lysine-specific demethylase 5A (KDM5A), also known as JARID1A, RBBP-2, or RBP2, is a histone demethylase that specifically removes methyl groups from trimethylated and dimethylated lysine 4 of histone H3 (H3K4), playing a pivotal role in chromatin regulation and gene transcription.
- KDM5A specifically demethylates H3K4me3 and H3K4me2 but not monomethylated H3K4, nor other histone lysine residues (H3K9, H3K27, H3K36, H3K79, H4K20), thereby contributing to histone code dynamics.
- It regulates gene transcription by binding directly to DNA at the 5'-CCGCCC-3' motif, impacting downstream target gene expression. Related references: PMID:18270511
- KDM5A is involved in transcriptional control of Hox proteins during cell differentiation, and may repress cytokines like CXCL12, as well as regulate circadian rhythm through a demethylase-independent coactivator function with CLOCK-BMAL1. Related references: PMID:19430464
- The protein localizes to the nucleus and nucleolus, consistent with its role in chromatin modification and transcription regulation.
- KDM5A has been reported to act as a transcriptional corepressor for certain genes (e.g., MT1F) and to favor the proliferation of cancer cells, highlighting its potential significance in oncology. Related references: PMID:27427228
- As a Jumonji/ARID domain-containing protein, KDM5A belongs to the JARID family of histone demethylases, which are characterized by their oxygenase activity requiring iron and 2-oxoglutarate.
Experimental Guidance and Technical Tips
- For Western blot (WB), given the large molecular weight of KDM5A (~192 kDa), use low-percentage acrylamide gels and ensure thorough protein transfer with appropriate buffers and extended transfer times.
- In immunoprecipitation (IP), the polyclonal nature of this antibody may enhance capture efficiency, but include isotype controls and validate recovery by target-specific detection.
- For chromatin immunoprecipitation (ChIP), this antibody is predicted to work based on the recombinant full-length immunogen; however, it is advisable to titrate the antibody and confirm enrichment of known KDM5A target promoters in your experimental system.
- ELISA applications may require optimization of coating antigen and antibody dilution; consider using the recombinant immunogen as a positive control.
