Biological Background: KDM4C Function and Localization

  • KDM4C, also known as JMJD2C or GASC1, is a member of the Jumonji C (JmjC) domain-containing histone demethylase family that specifically demethylates trimethylated histone H3 lysine 9 (H3K9me3) and lysine 36 (H3K36me3), but has no activity on mono- or dimethylated residues.
  • It does not demethylate histone H3 lysine 4 (H3K4) or lysine 27 (H3K27), nor histone H4 lysine 20 (H4K20), demonstrating its substrate specificity.
  • Demethylation by KDM4C produces formaldehyde and succinate as byproducts, requiring iron and α-ketoglutarate as cofactors.
  • The protein contains JmjN and JmjC domains, as well as zinc-finger motifs, which contribute to its chromatin targeting and catalytic activity.
  • KDM4C is primarily localized in the nucleus, consistent with its role in transcriptional regulation and chromatin remodeling.
  • Overexpression of KDM4C has been observed in several esophageal squamous cell carcinomas (ESCs), implicating it in cancer development.
  • Alternative splicing of the KDM4C gene generates multiple isoforms, though their functional differences are not fully characterized.

Experimental Guidance and Technical Tips

  • For Western blot analysis, detecting a band at approximately 120 kDa may require optimized reducing conditions and extended transfer times due to the high molecular weight.
  • When using this polyclonal antibody in ELISA, it can be employed as either a capturing or detecting antibody, though pairing should be validated with the specific assay setup.
  • Given the high sequence homology of KDM4C between human, mouse, and rat, this antibody is expected to cross-react; however, it is recommended to verify reactivity in your model system.
