Biological Background: KDM4B Function and Localization

  • KDM4B, also known as JMJD2B or JHDM3B, is a lysine-specific demethylase 4B and a member of the Jumonji C (JmjC) domain-containing histone demethylase family (gene KDM4B/JMJD2B). It is a chromatin-modifying enzyme that regulates transcription by removing methyl groups from histones.
  • Specifically demethylates trimethylated histone H3 lysine 9 (H3K9me3), producing formaldehyde and succinate in the process. It does not act on H3K4, H3K27, H3K36, or H4K20. Compared to other KDM4 family members (KDM4A, KDM4C, KDM4D), KDM4B has weaker catalytic activity toward H3K9me3. Related references: PMID:16603238 PMID:28262558
  • Plays a critical role in the development of the central nervous system (CNS), as highlighted by its implication in neurological functions and disease associations.
  • Localizes predominantly to the nucleus, consistent with its function in histone modification and chromatin regulation.
  • Contains multiple conserved domains including JmjC, JmjN, PHD-type zinc fingers, and TUDOR domains, which are involved in chromatin binding and catalytic activity. It requires iron and zinc as cofactors for its dioxygenase activity.
  • Subject to post-translational modifications, including acetylation and phosphorylation, which may regulate its activity or interactions. Alternative splicing generates multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 50-150 within the N-terminal region of human KDM4B. This region is distinct from the catalytic JmjC domain, and the antibody may recognize multiple isoforms that include this N-terminal sequence.
  • For Western blotting, a band near 122 kDa is expected. Validate specificity using appropriate positive and negative control lysates, and consider the possibility of detecting alternative splice variants.
  • For immunoprecipitation (IP) and chromatin immunoprecipitation (ChIP), perform pilot experiments to optimize antibody concentration and crosslinking conditions. Use IgG isotype controls and validate enrichment at known KDM4B target loci.
  • In immunofluorescence (IF/ICC), check nuclear localization; the antibody should predominantly stain the nucleus. Co-staining with nuclear markers can confirm specificity.
  • Cross-reactivity is reported for human, mouse, and rat. For other species, confirm sequence homology within the immunogen region before use.
