Biological Background: KDM4A Function and Localization

  • Lysine-specific demethylase 4A (KDM4A), also known as JMJD2A or JHDM3A, is a histone demethylase that specifically demethylates trimethylated 'Lys-9' (H3K9me3) and 'Lys-36' (H3K36me3) of histone H3, with no activity on mono- or dimethylated residues or on H3K4, H3K27, or H4K20. It also demethylates histone H1.4 at 'Lys-26' (H1.4K26me1/me2/me3).
    Related references: PMID:16603238, PMID:26741168, PMID:21768309, PMID:19144645, PMID:30156264
  • KDM4A participates in transcriptional repression by recruiting histone deacetylases (HDACs) and the co-repressor NCOR1 to E2F-responsive and ASCL2 promoters.
    Related references: PMID:16024779
  • A muscle-specific isoform of KDM4A lacking the N-terminal demethylase domain is crucial for muscle differentiation; it activates Myog gene transcription by removing repressive chromatin marks.
  • KDM4A is localized to the nucleus.
  • KDM4A is ubiquitously expressed across human tissues.
  • The protein belongs to the JmjC domain-containing histone demethylation family and contains zinc-finger domains. It is subject to acetylation, phosphorylation, and ubiquitination, and its activity involves iron and zinc as cofactors.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to residues 382-459 of human KDM4A, located in the N-terminal region. Verify that the antibody detects both full-length and muscle-specific isoforms lacking the N-terminal domain by testing in relevant cell lysates (e.g., myoblasts vs. myotubes).
  • For Western blot, a band at ~121 kDa is expected for full-length KDM4A; validate using nuclear extracts from human, mouse, or rat cell lines.
  • Optimize antibody dilution for each application (WB, IP, ELISA) using positive control samples; perform titration experiments to determine optimal working concentration.
  • For immunoprecipitation, consider cross-linking methods or direct protocols based on antibody isotype and target abundance.
  • Include appropriate controls (e.g., isotype control, knockout lysates) to confirm specificity.
