Biological Background: KDM2B Function and Localization
- KDM2B is a histone demethylase that specifically demethylates 'Lys-4' and 'Lys-36' of histone H3, with preference for trimethylated H3K4 and dimethylated H3K36.
Related references: PMID:16362057 PMID:17994099 PMID:26237645 - It binds to the transcribed region of ribosomal RNA and represses rRNA transcription, thereby inhibiting cell growth and proliferation.
Related references: PMID:16362057 PMID:17994099 - KDM2B may function as a substrate-recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex (probable).
- The protein localizes to the nucleus and nucleolus, and associates with chromosomes.
- KDM2B contains a CXXC zinc finger, a JmjC domain, an F-box, and leucine-rich repeats, contributing to its roles in chromatin regulation, DNA/RNA binding, and ubiquitin conjugation.
- Alternative splicing produces multiple isoforms, and mutations are associated with intellectual disability, as reflected in UniProt keywords.
- Post-translational modifications include phosphorylation and ubiquitination, as indicated by phosphoprotein and Ubl conjugation keywords.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 1059-1336) of human KDM2B. When validating antibody specificity, consider using a lysate from cells with high KDM2B expression or an overexpression control.
- For Western blot, the predicted molecular weight is approximately 153 kDa; ensure appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- In ELISA, use the recombinant protein as a positive control to establish a standard curve and assess linearity.
- Given KDM2B’s nuclear localization, nuclear extracts may be more suitable than whole-cell lysates for detection.
- Cross-reactivity is indicated for human; reactivity with other species should be verified experimentally if needed.
