Biological Background: KDM1A Function and Localization
- KDM1A (also known as LSD1, AOF2, BHC110) is a FAD-dependent lysine-specific histone demethylase that removes mono- and dimethyl marks from histone H3 at lysine 4 (H3K4me1/me2) and lysine 9 (H3K9me1/me2). Depending on the promoter context, this activity can lead to transcriptional repression or activation. Related references: PMID:15620353, PMID:15811342, PMID:16079794
- Demethylation of H3K4me on nucleosomes requires interaction with the co-repressor RCOR1/CoREST, linking KDM1A to the regulation of neuronal gene expression and REST complex function. Related references: PMID:16079794, PMID:16140033, PMID:16885027
- In the presence of androgen receptor (AR), KDM1A acts as a coactivator by demethylating H3K9me, and this function is modulated by phosphorylation of histone H3 threonine 6 (H3T6ph) by PRKCB. Related references: PMID:16079795
- KDM1A also demethylates non-histone substrates, including lysine 370 of p53 (impairing its interaction with 53BP1), DNA methyltransferase 1 (DNMT1), and the transcription factor SOX2, thereby influencing protein stability and function. Related references: PMID:29691401, PMID:29358331
- It plays a crucial role in development, being required for gastrulation, and in hematopoiesis where it acts within a RCOR/GFI/KDM1A/HDAC complex to regulate blood cell differentiation. Related references: PMID:16079794, PMID:16140033
- KDM1A facilitates epithelial-to-mesenchymal transition (EMT) by cooperating with SNAI1 to repress epithelial markers such as E-cadherin/CDH1, CDN7, and KRT8. Related references: PMID:20562920, PMID:27292636, PMID:20389281
- In neurons, a brain-enriched isoform of KDM1A demethylates histone H4 lysine 20 (H4K20me1/me2) to regulate activity-dependent gene expression, a process essential for spatial learning and memory. Related references: PMID:26214369
- KDM1A is predominantly localized to the nucleus and is ubiquitously expressed, with particularly high expression in brain and testis.
Experimental Guidance and Technical Tips
- For Western blot analysis, the predicted molecular weight of KDM1A is approximately 93 kDa; consider using appropriate molecular weight markers and verify band specificity with knockdown or knockout controls.
- The immunogen corresponds to residues 130–380 of human KDM1A; perform sequence alignment to assess target homology when using mouse or rat samples, as cross-reactivity is indicated but may vary by application.
- In immunohistochemistry (IHC-P), tissue sections from brain or testis may serve as positive controls given high KDM1A expression; optimize antigen retrieval and primary antibody dilution empirically.
- For immunofluorescence (IF/ICC), expect nuclear staining; proper cell fixation and permeabilization are critical, and co-staining with a nuclear marker is recommended.
- When used in ELISA, inclusion of positive and negative controls is essential to assess antibody specificity and linearity.
