  • KCNJ5 encodes the G protein-activated inward rectifier potassium channel 4 (also known as GIRK4, Kir3.4, CIR, KATP-1), a member of the inwardly rectifying potassium channel family.
  • It mediates potassium influx more readily than efflux, with voltage dependence modulated by extracellular potassium concentration; inward rectification primarily results from blockage of outward current by intracellular magnesium. The channel can be inhibited by external barium.
  • Channel activity is controlled by G proteins, linking it to G protein-coupled receptor signaling pathways.
  • The protein is localized to the plasma membrane (Membrane).
  • Expressed in pancreatic islets, exocrine pancreas, heart, and adrenal cortex (particularly zona glomerulosa), consistent with its roles in hormone secretion and cardiac excitability.
  • Classified as a phosphoprotein, suggesting regulation by phosphorylation.
  • Mutations in KCNJ5 are associated with long QT syndrome (LQT13) and other cardiac disorders (Disease variant).
