Biological Background: KCNIP2 Function and Localization
- KCNIP2, also known as KChIP2, is a regulatory subunit of Kv4/D (Shal)-type voltage-gated rapidly inactivating A-type potassium channels. It modulates channel density, inactivation kinetics, and rate of recovery from inactivation in a calcium-dependent and isoform-specific manner. Related references: PMID:10676964, PMID:11287421, PMID:11684073
- It facilitates trafficking of KCND2 and KCND3 potassium channel subunits to the cell surface. Related references: PMID:12829703
- KCNIP2 may be required for the expression of transient outward potassium currents (I(To)) in the heart (By similarity).
- Tissue expression: KCNIP2 is prominently expressed in brain, colocalizing with KCND2 in excitatory neurons including cortical and hippocampal CA1 pyramidal cells. Isoform 3 is additionally expressed in heart and umbilical vein endothelial cells, but not in fetal heart.
- Subcellular localization is primarily at the cell membrane, likely anchored via palmitoylation (as indicated by lipoprotein and palmitate keywords).
- The protein contains EF-hand calcium-binding motifs, classifying it as a neuronal calcium sensor subject to phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 1-90 of human KCNIP2) targets the N-terminal region—consider potential isoform specificity and verify using lysates from relevant tissues (e.g., brain or heart).
- For Western blot, expect a band around 31 kDa. Given indicated cross-reactivity with mouse and rat, the antibody may be suitable for comparative studies across species; validate in the appropriate sample system.
- For ELISA, ensure the coating antigen matches the immunogen region and optimize assay conditions empirically.
