Biological Background: KCNH2 Function and Localization
- KCNH2 (HERG/Kv11.1) is the pore-forming alpha subunit of a voltage-gated inwardly rectifying potassium channel characterized by slow deactivation and rapid inactivation kinetics, producing small outward currents during depolarization and large inward currents upon repolarization. - It forms a stable heteromeric complex with KCNE1 or KCNE2, which regulates its inward rectifier potassium channel activity. Related references: PMID:10219239 PMID:9230439
- Some KCNH2 isoforms lack intrinsic channel activity but modulate channel characteristics by forming heterotetramers, and these isoforms are retained intracellularly and degraded via the ubiquitin pathway.
- The channel localizes to the cell membrane.
- KCNH2 is highly expressed in the heart and brain, playing a critical role in cardiac repolarization and neuronal excitability. Isoforms such as USO are frequently overexpressed in cancer cells.
- Post-translational modifications include glycosylation, phosphorylation, and methylation, and mutations in KCNH2 are associated with long QT syndrome and short QT syndrome.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide corresponding to amino acids 850-950 of human KCNH2 (NP_000229.1). It is expected to recognize both native and denatured forms of the protein.
- For Western blotting, the predicted molecular weight is approximately 127 kDa. Use appropriate loading controls and validate in your sample system.
- For immunofluorescence and immunohistochemistry, consider using heart or brain tissue sections as positive controls, given the high expression in these tissues.
- The antibody cross-reacts with human and mouse, but performance in other species should be verified.
- As with any polyclonal antibody, lot-to-lot variability may occur; it is recommended to titrate the antibody for each application.
