Biological Background: KAT7 Function and Localization

  • Histone acetyltransferase KAT7 (also known as HBO1, HBOa, MYST2) catalyzes acetylation of histone H3 at Lys-14 (H3K14ac) and histone H4 at Lys-5, Lys-8, and Lys-12 (H4K5ac, H4K8ac, H4K12ac). H3K14ac regulates gene transcription, immune regulation, stem cell maintenance, and embryonic development; H4 acetylation promotes DNA replication licensing by facilitating chromatin loading of MCM complexes. Related references: PMID:16387653 PMID:21753189 PMID:24065767 PMID:10438470 PMID:19187766 PMID:20129055 PMID:11278932 PMID:18832067
  • KAT7-mediated H3K14ac enhances the processivity of RNA polymerase II during transcription, and is crucial for maintaining high expression of HOXA9 and HOXA10 in acute myeloid leukemia stem cells, supporting leukemia cell maintenance. Related references: PMID:31827282
  • In hematopoiesis, KAT7 forms a complex with BRD1/BRPF2 to direct H3K14ac, which is essential for erythroid differentiation; H3K14ac is also required for T-cell development. Related references: PMID:21753189
  • KAT7 localizes to centromeres, where it mediates histone acetylation to prevent SUV39H1-mediated centromere inactivation and acts as a positive regulator of CENPA assembly, ensuring centromere identity. Related references: PMID:27270040
  • Upon DNA damage, KAT7 is phosphorylated by ATR and recruited to UV-induced lesions, facilitating histone acetylation and XPC recruitment for efficient nucleotide excision repair. Related references: PMID:28719581
  • KAT7 inhibits NF-kappa-B signaling independently of its acetyltransferase activity, providing a non-catalytic regulatory function in immune and inflammatory responses. Related references: PMID:16997280
  • Subcellularly, KAT7 is predominantly nuclear, with enrichment at chromosomes and centromeres; it is also detected in the cytosol, suggesting dynamic localization.
  • KAT7 is ubiquitously expressed, with highest levels in testis, indicating a broad but specialized role in different tissues.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 55–193, an N-terminal region; consider validating the antibody’s specificity by comparing signals in wild-type versus KAT7-knockdown cell lysates.
  • For Western blotting, expect a band near 71 kDa; optimize blocking (e.g., 5% nonfat milk) and loading controls, and titrate the antibody dilution for your sample system.
  • In IHC-P, antigen retrieval (e.g., heat-induced epitope retrieval in citrate buffer) may be necessary; use testis sections as a positive control due to high KAT7 expression.
  • For IF/ICC, fix and permeabilize cells appropriately; KAT7 nuclear and centromeric staining can be confirmed with a DNA counterstain (e.g., DAPI).
  • ELISA assays can employ the recombinant immunogen as a standard to determine optimal coating concentration and antibody titer; cross-reactivity with mouse and rat orthologs allows multi-species assay development.
