Biological Background: KAT2B/PCAF Function and Localization
- KAT2B, also known as PCAF, P/CAF, or lysine acetyltransferase 2B, is a histone acetyltransferase of the GNAT family that plays a key role in transcriptional activation and chromatin remodeling.
- It acetylates histones H3 and H4 and nucleosome core particles, with a strong preference for acetylation at H3 lysine 9 (H3K9ac) (PubMed:8945521, PubMed:21131905).
Related references: PMID:8945521, PMID:21131905 - Beyond histones, KAT2B acetylates numerous non-histone substrates, including ACLY, MAPRE1/EB1, PLK4, RRP9/U3-55K, and TBX5, thereby influencing processes such as mitosis, centrosome duplication, and pre‑rRNA processing (PubMed:10675335, PubMed:23001180, PubMed:23932781, PubMed:27796307, PubMed:26867678, PubMed:29174768).
Related references: PMID:10675335, PMID:23001180, PMID:23932781, PMID:27796307, PMID:26867678, PMID:29174768 - KAT2B inhibits cell‑cycle progression and counteracts the mitogenic activity of the adenoviral oncoprotein E1A (PubMed:8684459). Additionally, it acts as a negative regulator of centrosome amplification by acetylating PLK4 (PubMed:27796307).
Related references: PMID:8684459, PMID:27796307 - As a circadian transcriptional coactivator, KAT2B enhances the activity of the NPAS2‑BMAL1 and CLOCK‑BMAL1 heterodimers, linking chromatin modification to the molecular clock (PubMed:14645221).
Related references: PMID:14645221 - KAT2B acetylates spermidine (PubMed:27389534) and is involved in heart and limb development through TBX5 acetylation, which regulates nucleocytoplasmic shuttling of TBX5 (PubMed:29174768).
Related references: PMID:27389534, PMID:29174768 - During HIV‑1 infection, KAT2B is recruited by the viral protein Tat and is thought to facilitate proviral chromatin remodeling.
- The protein localizes to the nucleus, cytoplasm, centrosome, microtubule organizing center, and cytoskeleton, reflecting its diverse functional roles.
- KAT2B is ubiquitously expressed, with highest levels in heart and skeletal muscle; it is also detected in skin keratinocytes (PubMed:20940255).

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N‑terminal 86 amino acids of human KAT2B. For Western blotting, expect a band at approximately 93 kDa. Validate the antibody in your specific sample system, as post‑translational modifications or splice variants may affect detection.
- For immunofluorescence/immunocytochemistry (IF/ICC), consider that KAT2B localizes to the nucleus, cytoplasm, and centrosomal regions. Include appropriate controls and permeabilization protocols to detect epitopes in these compartments.
- This polyclonal antibody has been reported to react with human, mouse, and rat samples; however, cross‑reactivity with other species should be tested individually.
- ELISAs can be developed using this antibody, but optimal coating and detection conditions must be established by the end‑user.
