Biological Background: KAT2A Function and Localization
- KAT2A is a versatile lysine acyltransferase that uses acetyl-CoA, succinyl-CoA, glutaryl-CoA, crotonyl-CoA, and lactoyl-CoA to modify histone and non-histone proteins, expanding its regulatory repertoire beyond classical acetylation. Related references: PMID:31542297 PMID:29211711 PMID:37040526 PMID:39561764
- In the SAGA and ATAC complexes, KAT2A functions as a histone acetyltransferase (HAT) with a strong preference for acetylating histone H3 at Lys-9 (H3K9ac), promoting epigenetic transcription activation. It also acetylates H1.4K34 and variant H2A.Z. Related references: PMID:17301242 PMID:19103755 PMID:29211711 PMID:21131905 PMID:22465951 PMID:29973595 PMID:31527837
- KAT2A mediates histone succinylation (H3K79succ) and glutarylation (H4K91glu), modifications that influence nucleosome stability and transcription. Succinylation depends on association with the 2-oxoglutarate dehydrogenase complex. Related references: PMID:29211711 PMID:31542297
- Beyond histones, KAT2A acetylates key regulatory proteins such as PPARGC1A (suppressing gluconeogenesis), TBX5 (controlling nucleocytoplasmic shuttling), and PLK4 (limiting centrosome amplification), linking its activity to metabolism, development, and cell cycle control. Related references: PMID:16753578 PMID:23142079 PMID:27796307 PMID:29174768
- KAT2A is critical for diverse processes including long-term memory consolidation, T-cell activation, craniofacial development, and embryonic stem cell pluripotency, acting through targeted gene expression programs.
- Subcellular localization includes the nucleus, chromosomes, cytoplasm, and centrosome, allowing KAT2A to regulate chromatin and cytoplasmic targets.
- Ubiquitously expressed across tissues, KAT2A also interacts with HIV-1 Tat protein to modulate proviral chromatin remodeling and transcriptional activity.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 1-100) of human KAT2A; thus, the antibody may recognize full-length KAT2A and certain splice variants. Validate specificity in your target tissue or cell line.
- For Western blotting, the predicted molecular weight is 94 kDa, but post-translational modifications (phosphorylation, acetylation) can cause band shifts. Consider using fresh protease/phosphatase inhibitors and optimize gel percentage accordingly.
- In IHC-P, ensure proper antigen retrieval (e.g., heat-induced epitope retrieval with citrate buffer pH 6.0) and validate staining with appropriate positive and negative controls. Cross-reactivity with mouse and rat makes this antibody suitable for multi-species studies, but always verify in the relevant sample system.
- For IF/ICC, permeabilization and fixation conditions should be optimized; KAT2A’s presence in both nuclear and cytoplasmic compartments may require careful imaging parameters.
- ELISA may be used for quantitative detection; titrate the antibody and pair with a suitable detection reagent. Since this is a polyclonal antibody, consider its potential for capturing multiple epitopes.
