- Encoded by the NCOA1 gene, KAT13A/SRC1 (also known as bHLHe74, RIP160) is a nuclear receptor coactivator and histone acetyltransferase.
- It directly binds and coactivates multiple nuclear receptors (e.g., PGR, GR, ER, RXRs, TRs, PPARs) in a hormone-dependent manner, stimulating transcription.
- Also serves as a coactivator for STAT3, STAT5A, STAT5B, and STAT6 transcription factors, linking cytokine signaling to gene expression.
- Possesses histone acetyltransferase (HAT) activity toward H3 and H4, potentially contributing to chromatin remodeling and transcription activation.
- Forms multisubunit coactivator complexes that participate in chromatin remodeling and recruitment of general transcription factors.
- Together with NCOA2, plays a role in controlling energy balance between white and brown adipose tissues.
- Isoforms 1, 2, and 3 differ in activity; isoform 2 exhibits the highest thyroid hormone-dependent transactivation.
