Biological Background: KAP1/TRIM28 Function and Localization

  • Also known as transcription intermediary factor 1-beta (TIF1-beta), KRAB-associated protein 1 (KAP-1), or KRIP-1, this 88.6 kDa tripartite motif (TRIM) family protein features RING, B-box, coiled-coil, PHD, and bromodomain regions.
  • Functions as an E3 SUMO and ubiquitin ligase that scaffolds transcriptional repression complexes by recruiting SETDB1 (histone methyltransferase) and NuRD (nucleosome remodeling and deacetylation) complex, leading to H3K9 methylation and heterochromatin formation.
  • Serves as a nuclear corepressor for KRAB domain-containing zinc finger proteins (KRAB-ZFPs), mediating silencing of endogenous retroviruses and genes critical for embryonic development and genomic imprinting.
