Biological Background: KAP1/TRIM28 Function and Localization

  • Transcription intermediary factor 1-beta (TIF1β), also known as KAP1/TRIM28, is an E3 SUMO and ubiquitin ligase essential for embryonic development, genomic imprinting, and maintaining genomic stability by silencing repetitive and retroviral elements.
  • Functions as a scaffold for transcriptional repression complexes, acting as a corepressor for KRAB-ZFPs by recruiting CHD3 (NuRD complex) and SETDB1 to establish H3K9me3 and heterochromatin.
  • Collaborates with SETDB1 to silence endogenous and introduced retroviruses via H3K9me3 in a DNA-methylation independent pathway (By similarity).
  • Plays a dual role as coactivator for CEBPB and NR3C1 and corepressor for ERBB4, and inhibits E2F1 activity by promoting E2F1-HDAC1 complex formation to suppress apoptosis.
