Biological Background: Kaiso/ZBTB33 Function and Localization
- Transcriptional regulator Kaiso, also known as Zinc finger and BTB domain-containing protein 33 (ZBTB33), encoded by the ZBTB33 gene (gene ID 10009).
- Exhibits bimodal DNA-binding specificity: binds to methylated CpG dinucleotides (consensus 5'-CGCG-3') and the non-methylated consensus sequence 5'-CTGCNA-3' (kaiso binding site, KBS).
- Recruits the N-CoR repressor complex to promote histone deacetylation and formation of repressive chromatin structures at target gene promoters, thereby regulating transcription.
- May contribute to the repression of target genes of the Wnt signaling pathway, and can also activate transcription of a subset of target genes by recruiting CTNND2.
- Represses expression of MMP7 in conjunction with transcriptional corepressors CBFA2T3, CBFA2T2, and RUNX1T1. Related references: PMID:23251453
- Subcellular localization is primarily nuclear but also detected in the cytoplasm.
- Tissue specificity: expressed in vascular endothelium.
