  • Transcriptional regulator Kaiso (ZBTB33/ZNF348) functions as a bimodal DNA-binding protein, recognizing methylated CpG dinucleotides (5'-CGCG-3') and the non-methylated Kaiso binding site (5'-CTGCNA-3').
  • Recruits the N-CoR corepressor complex to target gene promoters, promoting histone deacetylation and repressive chromatin formation, thereby negatively regulating genes of the Wnt signaling pathway.
  • May also act as a transcriptional activator for a subset of target genes via recruitment of CTNND2 (catenin delta-2), demonstrating context-dependent regulatory roles.

For quantitative applications, validate antibody specificity in your relevant sample system.
  • For Western blotting, a band around 74 kDa is expected. Include positive controls from human, mouse, or rat cell lysates if available; consider that endogenous expression may vary by tissue or cell type.
  • In chromatin immunoprecipitation (ChIP), optimize fixation and chromatin shearing conditions, as Kaiso binds to methylated DNA; the antibody may be suitable for both ChIP-qPCR and ChIP-seq, but confirm binding in your system.
