Biological Background: Kaiso/ZBTB33 Function and Localization
- Transcriptional regulator Kaiso (ZBTB33/ZNF348) functions as a bimodal DNA-binding protein, recognizing methylated CpG dinucleotides (5'-CGCG-3') and the non-methylated Kaiso binding site (5'-CTGCNA-3').
- Recruits the N-CoR corepressor complex to target gene promoters, promoting histone deacetylation and repressive chromatin formation, thereby negatively regulating genes of the Wnt signaling pathway.
- May also act as a transcriptional activator for a subset of target genes via recruitment of CTNND2 (catenin delta-2), demonstrating context-dependent regulatory roles.
- Represses matrix metalloproteinase 7 (MMP7) expression through interactions with corepressors CBFA2T3, CBFA2T2, and RUNX1T1, as shown in functional studies (PubMed:23251453). Related references: PMID:23251453
- Subcellularly located in both the nucleus and cytoplasm, consistent with its roles in transcriptional regulation and potential shuttling.

- The immunogen corresponds to the C-terminal region (aa 523-672) of human Kaiso, which contains zinc finger domains; this region is well conserved across human, mouse, and rat, consistent with the reported cross-reactivity. For quantitative applications, validate antibody specificity in your relevant sample system.
- For Western blotting, a band around 74 kDa is expected. Include positive controls from human, mouse, or rat cell lysates if available; consider that endogenous expression may vary by tissue or cell type.
- In chromatin immunoprecipitation (ChIP), optimize fixation and chromatin shearing conditions, as Kaiso binds to methylated DNA; the antibody may be suitable for both ChIP-qPCR and ChIP-seq, but confirm binding in your system.
- For ELISA, use the recombinant immunogen as a standard to establish assay conditions; note that the polyclonal nature may require careful optimization of coating and detection antibodies.
