Biological Background: Kaiso Function and Localization

  • Also known as zinc finger and BTB domain-containing protein 33 (ZNF348/ZNF-kaiso), Kaiso is a transcriptional regulator encoded by the ZBTB33 gene in human.
  • Functions as a bimodal DNA-binding protein: it recognizes methylated CpG dinucleotides in the consensus 5'-CGCG-3' sequence and the non-methylated kaiso binding site (KBS) 5'-CTGCNA-3'.
  • Recruits the N-CoR repressor complex to gene promoters, promoting histone deacetylation and formation of repressive chromatin structures to silence target genes of the Wnt signaling pathway.
  • May also activate transcription of certain target genes by recruiting CTNND2, demonstrating context-dependent regulatory duality.
  • Represses MMP7 expression through cooperation with corepressors CBFA2T3, CBFA2T2 and RUNX1T1. Related references: PMID:23251453
  • Primarily localizes to the nucleus, with cytoplasmic distribution also observed, consistent with its roles in transcriptional regulation.
  • Expressed in vascular endothelium and implicated in pathways involving DNA binding, metal (zinc) coordination, and post-translational modifications such as phosphorylation and ubiquitination.
