Biological Background: JunD Function and Localization
- Transcription factor JunD is a member of the Jun family of basic leucine zipper (bZIP) transcription factors and functions as a key component of the AP-1 transcription complex.
- JunD binds specifically to AP-1 consensus sequences (5'-TGA[GC]TCA-3') within regulatory regions of target genes, regulating their expression (PubMed:9989505). Related references: PMID:9989505
- JunD heterodimerizes with proteins of the FOS family to form the AP-1 transcription factor complex, enhancing both DNA-binding affinity and transcriptional activity (PubMed:28981703, PubMed:9989505). Related references: PMID:9989505, PMID:28981703
- JunD is predominantly localized in the nucleus, consistent with its role in transcriptional regulation.
- Post-translational modifications, including phosphorylation, modulate JunD activity and protein interactions.
- Additional relevant keywords include DNA-binding, Activator, Disulfide bond, and Proteomics identification.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 100–200 of human JunD; this antibody likely recognizes the native protein in both denaturing and native conditions.
- For Western blotting, validate the antibody concentration in your sample system; a band near 35 kDa is expected under reducing conditions.
- In ChIP assays, optimize cross-linking and shearing conditions to enrich JunD-bound DNA fragments; include appropriate positive and negative controls.
- For CUT&Tag experiments, follow the manufacturer’s protocol for target-specific antibody titration to ensure high-resolution genome-wide profiling.
- In ELISA applications, this antibody can serve as either a capture or detection reagent; verify optimal working concentrations by serial dilution.
