Biological Background: JNK1/2/3 Function and Localization

  • MAPK8 (JNK1) is a serine/threonine-protein kinase that belongs to the mitogen-activated protein kinase family. Together with MAPK9 (JNK2) and MAPK10 (JNK3), it forms the c-Jun N-terminal kinase (JNK) subfamily.
  • JNK1 plays a central role in the stress-activated protein kinase/c-Jun N-terminal kinase (SAP/JNK) signaling pathway, responding to extracellular stimuli such as pro-inflammatory cytokines or physical stress. It is activated by dual phosphorylation by MAP2K4/MKK4 and MAP2K7/MKK7. Related references: PMID:28943315
  • Activated JNK1 phosphorylates various transcription factors, primarily components of AP-1 such as JUN, JDP2, and ATF2, thereby regulating AP-1 transcriptional activity. Related references: PMID:18307971
  • JNK1 is involved in diverse cellular processes including cell proliferation, differentiation, migration, transformation, and programmed cell death. It promotes apoptosis by phosphorylating key regulators like p53/TP53 and YAP1. Related references: PMID:21364637
  • Subcellularly, JNK1 is found in the cytoplasm and nucleus, and also localizes to synapses in neurons.
  • JNK1 phosphorylates a wide range of substrates, including CDT1 (involved in replication licensing), BCL2 (mediating autophagy), HSF1, SIRT1, ELK1, ITCH, and NLRP3, linking it to processes such as DNA replication, autophagy, circadian rhythm, and inflammasome activation. Related references: PMID:21856198, PMID:18570871
  • Keywords associated with JNK1 include alternative splicing, ATP-binding, biological rhythms, kinase, phosphoprotein, and reference proteome, indicating its regulatory complexity and importance.

Experimental Guidance and Technical Tips

  • This polyclonal antibody, raised against a conserved region of JNK1, is expected to recognize JNK1, JNK2, and JNK3. In Western blotting, multiple bands corresponding to different isoforms and splice variants may be observed (approx. 27–52 kDa), so verify the specific band pattern in your sample.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization conditions may need optimization; consider testing a dilution series.
  • In immunoprecipitation (IP) experiments, use a suitable amount of antibody per sample and confirm capture by subsequent Western blotting.
  • This antibody can be applied in ELISA for detecting JNK proteins in cell lysates or recombinant preparations; include negative controls to assess specificity.
