Biological Background: JNK1 Function and Localization
- Mitogen-activated protein kinase 8 (JNK1), encoded by the MAPK8 gene, is a stress-activated serine/threonine kinase that functions as a key component of the c-Jun N-terminal kinase (JNK) signaling cascade. It is also known as SAPK1c and JNK-46.
- Upon activation by dual phosphorylation from upstream kinases MAP2K4/MKK4 and MAP2K7/MKK7, JNK1 translocates to the nucleus and phosphorylates transcription factors such as c-Jun and ATF2, thereby regulating AP-1-dependent gene expression. Related references: PMID:18307971
- JNK1 promotes stress-induced apoptosis by phosphorylating key proteins including p53 and YAP1, linking cellular stress to programmed cell death. Related references: PMID:21364637

Experimental Guidance and Technical Tips
- The immunogen was designed against residues 245–345 within the kinase domain of human JNK1, a region highly conserved among species, which supports the reported cross-reactivity with mouse and rat. Researchers may consider verifying reactivity in their specific model system.
- For Western blotting, a predominant band around 48 kDa is expected. Include appropriate positive and negative controls; whole-cell lysates from stressed or stimulated cells (e.g., UV-treated HeLa) may enhance signal due to activation-induced stabilization.
- In immunocytochemistry/immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), both nuclear and cytoplasmic staining patterns are anticipated. For neuronal tissues, synaptic localization may also be observed. Optimize fixation and permeabilization conditions for each sample type.
- Because this antibody recognizes total JNK1, detection of the activated (phosphorylated) form requires separate phospho-specific antibodies. It is recommended to run parallel blots or multiplex staining to distinguish activation states.
- For ELISA applications, consider using the antibody as a capture or detection reagent against recombinant JNK1 or cell lysates, pairing with a non-competing antibody for sandwich formats.
