Biological Background: JMJD7 Function and Localization
- JMJD7 is a bifunctional enzyme possessing both endopeptidase and 2-oxoglutarate-dependent monooxygenase activities. Related references: PMID:28847961 PMID:29915238
- It cleaves the N-terminal tails of histones H2, H3, and H4 at the carboxyl side of methylated arginine or lysine residues, generating 'tailless nucleosomes' that may facilitate transcription elongation. Related references: PMID:28847961
- JMJD7 preferentially recognizes and cleaves monomethylated and dimethylated arginine residues, and can further digest histone tails via its aminopeptidase activity. Related references: PMID:28847961
- As a (3S)-lysyl hydroxylase, JMJD7 catalyzes Fe(2+) and 2-oxoglutarate-dependent stereospecific hydroxylation of 'Lys-22' in DRG1 and 'Lys-21' in DRG2 translation factors, potentially modulating their interaction with ribonucleic acids. Related references: PMID:29915238
- The protein localizes to both the nucleus and cytoplasm, consistent with its dual roles in chromatin modification and translation regulation.
- Structurally, JMJD7 contains a JmjC domain, a disulfide bond, and metal-binding sites, and is classified under hydrolases, oxidoreductases, and proteases.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the full-length human JMJD7 (aa 1-316). Due to high sequence homology among human, mouse, and rat, this antibody is expected to cross-react with these species, but independent validation is recommended.
- For Western blotting, a specific band around 36 kDa is anticipated. Use appropriate nuclear and cytoplasmic extraction protocols to detect JMJD7 in both compartments.
- ELISA performance may vary depending on coating antigen and detection format. Optimization of coating concentration and antibody dilution is advised.
- Consider including appropriate controls (e.g., recombinant protein, knockout cell lysates) to confirm specificity in your experimental system.
