Biological Background: JARID2 Function and Localization
- JARID2, also known as Jumonji/ARID domain-containing protein 2, is a 139 kDa nuclear protein belonging to the Jumonji family, but it lacks histone demethylase activity.
- It functions as a regulator of histone methyltransferase complexes, playing an essential role in embryonic development, including heart and liver development, neural tube fusion, and hematopoiesis. Related references: PMID:20075857
- Acts as an accessory subunit of the PRC2 (Polycomb repressive complex 2) complex, which mediates histone H3K27 trimethylation (H3K27me3) on chromatin, a repressive mark critical for gene silencing. Related references: PMID:20075857, PMID:29499137, PMID:31959557

- For Western blot, a band around 139 kDa is expected. Use appropriate gel percentage and transfer conditions for high molecular weight proteins. Validate detection in nuclear extracts due to predominant nuclear localization.
