Biological Background: JAM-A Function and Localization
- JAM-A (junctional adhesion molecule A), also called JAM-1 or F11 receptor, is an immunoglobulin superfamily protein that mediates homophilic cell adhesion and regulates tight junction assembly. Experimental Guidance and Technical Tips
- For Western blotting, a band at approximately 32 kDa is expected in human, mouse, and rat lysates. Consider running appropriate positive and negative controls, and validate expression in your target tissue or cell line.
- In immunofluorescence/ICC, JAM-A should exhibit membrane and junctional staining. Optimize fixation and permeabilization conditions, as tight junction proteins can be sensitive to over-fixation.
- For ELISA applications, the antibody can be used as a capture or detection reagent. If developing a sandwich ELISA, pair with a validated matched antibody to ensure specificity and sensitivity.
- The recombinant protein immunogen suggests the antibody may recognize both native and denatured epitopes, but empirical validation in each assay format is recommended.
