Biological Background: JAK3 Function and Localization
- JAK3 (Janus kinase 3) is a non-receptor tyrosine-protein kinase encoded by the JAK3 gene in humans. Alternative splicing produces two isoforms: the longer S-form and the shorter B-form, which differ in tissue distribution.
- It plays a central role in cytokine signal transduction by associating with type I receptors that share the common gamma chain (IL2RG), including receptors for IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21.
- Upon ligand binding, JAK3 phosphorylates tyrosine residues on the receptor cytoplasmic tail, creating docking sites for STAT transcription factors; it then phosphorylates STATs (e.g., STAT5A/B), which dimerize and translocate to the nucleus to regulate gene expression.
- JAK3 is essential for both innate and adaptive immunity, and is critically involved in T-cell development and hematopoiesis.
- Subcellularly, JAK3 is localized to the cytoplasm and is associated with the endomembrane system.
- Tissue expression: JAK3 is detected in NK cells and NK-like cell lines, but not in resting T-cells. The S-form predominates in hematopoietic lineages, while the B-form is found in both hematopoietic and epithelial cells.

- When performing ELISA, use appropriate blocking and detection systems; pairing with another JAK3 antibody may be beneficial for sandwich assays.
- As a polyclonal antibody, lot-to-lot variability may affect performance; validation in the relevant sample system is recommended.
- Note that JAK3 is primarily expressed in immune cells; positive controls could include NK cell lines or activated T-cells.
