Biological Background: JAK1 Function and Localization
- JAK1 (JTK3; JAK1A) is a 133 kDa non-receptor tyrosine kinase that belongs to the Janus kinase family and contains SH2 and kinase domains, along with tyrosine-protein kinase and transferase activities.
- It phosphorylates and activates signal-transducing subunits of various cytokine receptors, including IL-2, IL-10, IL-6, IL-31, IFN-alpha/beta/gamma, and oncostatin-M receptor complexes, thereby initiating STAT protein recruitment and signaling. - Upon interferon binding, JAK1 phosphorylates IFNAR2, creating docking sites for STAT proteins that then become phosphorylated and translocate to the nucleus to activate transcription. Related references: PMID:7759950, PMID:16239216, PMID:32750333, PMID:8232552
- Beyond interferon signaling, JAK1 is necessary for humanin/MT-RNR2-driven STAT3 phosphorylation, CNTF/CLCF1-induced STAT3 activation via LIFR/IL6ST, oncostatin-M signaling through type I and II receptors, and Interleukin-31 signaling through the OSMR/IL31RA complex. Related references: PMID:27384491, PMID:11294841, PMID:9188471, PMID:15194700
- Subcellularly, JAK1 is associated with the endomembrane system, consistent with its role in cytokine receptor signaling at the plasma membrane and intracellular compartments.
- Post-translational modifications include acetylation and ubiquitination (Ubl conjugation); the kinase domain requires ATP and metal ions (Mg²⁺) for activity, and the protein features an SH2 domain and phosphotyrosine residues.
- Tissue specificity data indicate higher expression in primary colon tumors than in normal colon tissue, with metastatic tumors showing expression levels comparable to normal tissue.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to aa 600-700 of human JAK1 (P23458), a region that may be part of the pseudokinase domain or interdomain linker; consider verifying the antibody specificity by peptide competition or knockout validation.
- For Western blot, the predicted molecular weight is ~133 kDa; optimize loading controls and blocking conditions to minimize non-specific binding, especially in immunoprecipitation or complex lysates.
- In ELISA, ensure the coating antigen presents the epitope effectively; test a range of antibody dilutions to determine optimal signal-to-noise ratio.
- Cross-reactivity with mouse and rat is anticipated based on supplier data, but validation in species-specific cell or tissue lysates is advised.
