Biological Background: JADE1 Function and Localization
- JADE1 (Jade family PHD finger protein 1), also known as PHF17, is a scaffold subunit of HBO1 histone acetyltransferase complexes that specifically acetylate histone H4 at lysines 5, 8, and 12 (H4K5ac, H4K8ac, H4K12ac), regulating DNA replication initiation.
Related references: PMID:16387653, PMID:19187766, PMID:20129055, PMID:24065767 - JADE1 directs KAT7/HBO1 substrate specificity toward histone H4 rather than other histones, playing a critical role in chromatin remodeling and transcriptional control.
Related references: PMID:20129055, PMID:24065767 - Beyond histone acetylation, JADE1 interacts with KAT5 to promote acetylation of nucleosomal histone H4, and it functions in apoptosis pathways, potentially acting as a renal tumor suppressor.
Related references: PMID:15502158, PMID:16046545 - JADE1 negatively regulates canonical Wnt signaling, in part through cooperation with NPHP4, linking it to developmental and kidney disease pathways.
Related references: PMID:22654112 - The protein contains PHD finger domains (zinc-finger motifs) that mediate chromatin binding, and it is subject to post-translational modifications including acetylation, phosphorylation, and ubiquitination.
- JADE1 localizes primarily to the nucleus and chromosomes, but is also detected in the cytoplasm, cytoskeleton, and at the cilium basal body, indicating dynamic subcellular distribution.
- Tissue expression is highest in kidney, with detectable levels in pancreas, liver, and heart; downregulation in renal cancer cells supports a tumor suppressor role.
Experimental Guidance and Technical Tips
- The immunogen encompasses a central region (aa 210-509) of human JADE1, which contains functional domains critical for its interactions; this design may capture isoforms and ensure detection in denaturing and native conditions.
- For Western blotting (WB), use standard reducing conditions and load 20-50 μg of total protein lysate from target-expressing cells/tissues; expect a band around 96 kDa. Include positive (e.g., kidney lysate) and negative controls.
- In immunofluorescence (IF/ICC), fix/permeabilize cells and use an appropriate dilution; JADE1 localizes to the nucleus and cytoplasm, so DAPI co-staining is recommended to confirm nuclear signal.
- For ELISA applications, the antibody can be used as capture or detection reagent; titrate and validate with recombinant JADE1 protein or peptide to establish optimal working concentrations.
