Biological Background: Intersectin-2 Function and Localization
- ITSN2 (intersectin-2) is a multidomain adapter protein also known as SH3P18 or SWAP, with a molecular weight of approximately 193 kDa.
- It contains SH3 domains, a coiled-coil region, and calcium-binding motifs, and belongs to the intersectin family of endocytic scaffold proteins.
- ITSN2 provides an indirect link between endocytic membrane traffic and the actin assembly machinery, regulating the formation and maturation of clathrin-coated vesicles (CCVs).
- Plays a role in endocytosis of integrin beta-1 (ITGB1) and transferrin receptor (TFR), facilitating cargo internalization.
- Involved in dendrite formation by melanocytes, as evidenced by functional studies. Related references: PMID:23999003
- Localizes to the cytoplasm, consistent with its role in endocytic trafficking.
- Expressed ubiquitously; isoform 1 is predominantly expressed in adult heart and liver, and it is also expressed in melanocytes.
