Biological Background: ITPR3 Function and Localization

  • ITPR3 encodes inositol 1,4,5-trisphosphate receptor type 3 (IP3R-3), a member of the inositol 1,4,5-trisphosphate receptor (IP3R) family of intracellular calcium release channels.
  • The ITPR3 protein functions as an IP3-gated calcium channel, releasing Ca²⁺ from the endoplasmic reticulum into the cytoplasm upon IP3 binding, thereby regulating cellular calcium homeostasis.
    Related references: PMID:8081734, PMID:32949214, PMID:37898605
  • Structural studies reveal a gating mechanism where IP3 binding stabilizes a preactivated state, enhancing calcium sensitivity; calcium binding at the juxtamembrane domain then promotes channel opening, while high cytosolic calcium later binds to a cytoplasmic domain to inhibit the channel.
    Related references: PMID:30013099, PMID:35301323, PMID:37898605
  • ITPR3 localizes primarily to the endoplasmic reticulum membrane and is also found in cytoplasmic vesicles and secretory vesicle membranes.
  • Tissue expression of ITPR3 is prominent in intestinal crypt and villus epithelial cells.
  • Mutations in ITPR3 are associated with Charcot-Marie-Tooth disease and ectodermal dysplasia, highlighting its clinical significance.
  • The protein is a phosphoprotein with a large molecular weight of approximately 304 kDa.

Experimental Guidance and Technical Tips

  • Given the large protein weight (304 kDa), consider using a low-percentage SDS-PAGE gel (e.g., 6%) and extended transfer times for Western blotting.
  • For immunofluorescence (IF/ICC), permeabilization with detergents such as Triton X-100 or digitonin may be required to allow antibody access to the endoplasmic reticulum.
  • The immunogen spanning amino acids 1630–1870 targets the C-terminal region of ITPR3, which may be useful for detecting full-length protein or specific fragments.
  • Validate the antibody in your specific sample system; optimal working dilutions for each application should be determined experimentally.
