Biological Background: ITLN1 Function and Localization
- ITLN1 is a calcium-dependent lectin that specifically recognizes microbial carbohydrate chains, binding to terminal acyclic 1,2-diol moieties such as β-linked D-galactofuranose (β-Galf), D-phosphoglycerol-modified glycans, KO and KDO. Related references: PMID:11313366 PMID:26148048
- It binds glycans from both Gram-positive and Gram-negative bacteria, including K. pneumoniae, S. pneumoniae, Y. pestis, P. mirabilis, and P. vulgaris. Related references: PMID:26148048
- ITLN1 does not bind human glycans, indicating selective microbial recognition. Related references: PMID:26148048
- Probably plays a role in the defense system against microorganisms.
- May act as an adipokine, enhancing insulin-stimulated glucose uptake in adipocytes without affecting basal uptake, and increases AKT phosphorylation in the absence and presence of insulin. Related references: PMID:16531507
- May interact with lactoferrin/LTF to increase its uptake, potentially contributing to iron absorption. Related references: PMID:11747454 PMID:23921499
- Located at the cell membrane and also secreted; contains a GPI-anchor, disulfide bonds, and glycosylation.
- Highly expressed in omental adipose tissue (stromal vascular cells) and small intestine; also detected in heart, testis, colon, salivary gland, skeletal muscle, pancreas, thyroid, and at lower levels in uterus, spleen, prostate, lymph node, and thymus. Related references: PMID:16531507
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight is 36 kDa; consider using reducing SDS-PAGE conditions and validate with positive control lysates from omental adipose tissue or small intestine.
- For immunohistochemistry (IHC-P), antigen retrieval may be necessary; validate staining in paraffin-embedded omental adipose tissue or intestinal sections where ITLN1 is highly expressed.
- This rabbit polyclonal antibody recognizes multiple epitopes on the immunogen, which may yield robust signals; however, titration and optimization are recommended for each application.
- Cross-reactivity with mouse and rat ITLN1 suggests utility in rodent model studies; confirm specificity in the relevant species before quantitative analysis.
- Because ITLN1 is both membrane-bound and secreted, consider sample preparation strategies that preserve both forms when designing experiments.
