Biological Background: ITK/EMT Function and Localization

  • Tyrosine-protein kinase ITK/TSK, also known as interleukin-2-inducible T-cell kinase, kinase EMT, T-cell-specific kinase, or tyrosine-protein kinase Lyk, is encoded by the ITK gene.
  • Plays an essential role in adaptive immunity by regulating the development, function, and differentiation of conventional T-cells and nonconventional NKT-cells. Upon TCR engagement, ITK is recruited to the membrane, phosphorylated by LCK, and autophosphorylates to become fully active.
  • Once activated, ITK phosphorylates PLCG1, leading to calcium release and NFAT nuclear translocation, and phosphorylates adapter proteins LAT and LCP2, ultimately driving lymphokine production, T-cell proliferation, and differentiation.   • Localizes to both the cytoplasm and nucleus.
  • Expressed in T-cell lines and natural killer cell lines.
  • Phosphorylates TBX21 at Tyr-530, mediating its interaction with GATA3; also involved in gamma-delta T-cell calcium response and transcriptomic modulation.
  • Contains SH2, SH3, and kinase domains with a zinc-finger motif; classified as a tyrosine-protein kinase with ATP-binding and metal-binding properties.

Experimental Guidance and Technical Tips

  • For Western blot, consider using whole cell lysates from human T-cell lines (e.g., Jurkat) to detect the ~72 kDa band.
  • For ELISA, validate with recombinant ITK protein or cell lysates to assess binding specificity; include appropriate positive and negative controls.
  • Given the immunogen is full-length recombinant protein, the antibody may recognize multiple epitopes; confirm specificity using knockout or knockdown models.
  • Ensure sample preparation includes protease and phosphatase inhibitors to preserve phosphorylation states and protein integrity.
