Biological Background: Integrin beta-6 Function and Localization
- ITGB6 encodes integrin beta-6, which associates with alpha-V to form the heterodimeric receptor integrin alpha-V:beta-6 (ITGAV:ITGB6).
- ITGAV:ITGB6 serves as a receptor for fibronectin and cytotactin, recognizing the R-G-D motif. Its clathrin-mediated internalization promotes carcinoma cell invasion. Related references: PMID:17158881, PMID:17545607
- This integrin mediates R-G-D-dependent release and activation of latent TGF-beta-1 from LAP, a critical step in TGF-beta signaling. Related references: PMID:15184403, PMID:22278742, PMID:28117447
- ITGAV:ITGB6 also acts as a receptor for fibrillin-1 (FBN1) and supports R-G-D-dependent cell adhesion to FBN1. Related references: PMID:17158881
- In microbial infection, ITGAV:ITGB6 acts as a host receptor for Coxsackievirus A9, Coxsackievirus B1, and Herpes simplex virus-1 (HHV-1). Related references: PMID:24367260
- Subcellularly, the protein localizes to the cell membrane and focal adhesions, functioning as a transmembrane adhesion receptor.
- Key structural features include EGF-like domains, disulfide bonds, and N-glycosylation sites; it requires divalent cations (Ca²⁺, Mg²⁺) for activity.
- Alternative splicing yields multiple isoforms, and mutations in ITGB6 are associated with amelogenesis imperfecta.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the cytoplasmic tail (aa 732–788) of human ITGB6; as a rabbit polyclonal, epitope recognition may vary across isoforms and species, so validation in the target sample is recommended.
- For Western blot, a band of approximately 86 kDa is expected; due to glycosylation, the observed molecular weight may be slightly higher. Use reducing conditions and appropriate membrane protein extraction protocols.
- For ELISA, recombinant ITGB6 protein or cell lysates from ITGB6-expressing lines can be used as coating antigens; cross-reactivity with human, mouse, and rat allows multi-species assay development.
- The antibody is supplied in liquid form, shipped on ice bags; upon receipt, store at -20°C and avoid repeated freeze-thaw cycles.
