- ITGB3BP (CENPR, NRIF3) is a multifunctional transcription coregulator that can act as either a coactivator or corepressor, and also functions as a component of the centromere/kinetochore complex.
- Isoform 1 specifically coactivates nuclear receptors for retinoid X (RXRs) and thyroid hormone (TRs) in a ligand-dependent manner, as well as estrogen receptor alpha, but does not coactivate receptors for retinoic acid, vitamin D, progesterone, or glucocorticoids.
- Acts as a transcriptional corepressor through interaction with the NFKB1 NF-kappa-B subunit, likely by interfering with its transactivation domain.
- Induces apoptosis selectively in breast cancer cells via a caspase-2-mediated pathway that involves mitochondrial membrane permeabilization but does not require other caspases.
- May function as an inhibitor of cyclin A-associated kinase activity.
- As a member of the CENPA-CAD (nucleosome distal) complex, it is recruited to centromeres and is essential for kinetochore protein assembly, proper mitotic progression, and chromosome segregation.
