Biological Background: ITGA3 Function and Localization

• Integrin alpha-3 (ITGA3) is a transmembrane glycoprotein that pairs with integrin beta-1 to form the VLA-3 receptor complex. It is also known as CD49C, FRP-2, GAPB3, and VLA-3 subunit alpha, with multiple isoforms generated by alternative splicing.
• As part of integrin alpha-3/beta-1, it functions as a receptor for extracellular matrix proteins including fibronectin, laminin, collagen, epiligrin, thrombospondin, and CSPG4, mediating cell adhesion and signaling.
• The alpha-3/beta-1 integrin provides a docking site for FAP (seprase) at invadopodia plasma membranes in a collagen-dependent manner, thereby facilitating adhesion, invadopodia formation, matrix degradation, and cell invasion.
• Together with LGALS3, it mediates CSPG4-stimulated endothelial cell migration, highlighting a role in angiogenesis.
