Biological Background: ITGA3 Function and Localization
- Integrin alpha-3 (ITGA3) is a transmembrane glycoprotein that pairs with integrin beta-1 to form the VLA-3 receptor complex. It is also known as CD49C, FRP-2, GAPB3, and VLA-3 subunit alpha, with multiple isoforms generated by alternative splicing.
- As part of integrin alpha-3/beta-1, it functions as a receptor for extracellular matrix proteins including fibronectin, laminin, collagen, epiligrin, thrombospondin, and CSPG4, mediating cell adhesion and signaling.
- The alpha-3/beta-1 integrin provides a docking site for FAP (seprase) at invadopodia plasma membranes in a collagen-dependent manner, thereby facilitating adhesion, invadopodia formation, matrix degradation, and cell invasion.
- Together with LGALS3, it mediates CSPG4-stimulated endothelial cell migration, highlighting a role in angiogenesis.
- ITGA3:ITGB1 may serve as a receptor for Rhizopus delemar CotH7 in alveolar epithelial cells, potentially contributing to pulmonary mucormycosis pathogenesis.
- Subcellularly, ITGA3 localizes to the cell membrane and is enriched in cell projections, including invadopodium and filopodium membranes, consistent with its roles in adhesion and migration.
- Post-translational modifications include palmitoylation, phosphorylation, disulfide bond formation, and cleavage on basic residues; it also binds calcium and other metals, features critical for integrin activation and function.
