Biological Background: ITCH Function and Localization
- ITCH (E3 ubiquitin-protein ligase Itchy homolog), also known as AIP4, NAPP1, is a HECT-type E3 ubiquitin ligase.
- Acts as an E3 ubiquitin-protein ligase which accepts ubiquitin from an E2 ubiquitin-conjugating enzyme and transfers it to targeted substrates. Related references: PMID:11046148, PMID:14602072, PMID:15051726
- Catalyzes 'Lys-29'-, 'Lys-48'- and 'Lys-63'-linked ubiquitin conjugation, enabling diverse signaling outcomes. Related references: PMID:17028573, PMID:18718448, PMID:19131965
- Essential component of a ubiquitin-editing protein complex (A20 complex) that ensures the transient nature of inflammatory signaling by promoting RIPK1 degradation. Related references: PMID:19131965
- Negatively regulates MAVS-dependent antiviral responses by ubiquitinating MAVS through 'Lys-48'-linked conjugation, leading to its proteasomal degradation. Related references: PMID:19881509
- Regulates endosomal sorting and degradation of receptors such as CXCR4 and FZD4 via ubiquitination of ESCRT-0 components and PI4K2A. Related references: PMID:14602072, PMID:23146885, PMID:34927784
- Together with UBR5, regulates apoptosis and reactive oxygen species levels through 'Lys-48'-/'Lys-63'-branched ubiquitination and proteasomal degradation of TXNIP. Related references: PMID:20068034, PMID:29378950
- Localizes to the cell membrane, cytoplasm, nucleus, and endosome membranes.
- Widely expressed across tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–100 of human ITCH, so the antibody likely recognizes the N-terminal region. Confirm specificity using knockout cell lysates or peptide competition assays.
- For WB, the predicted molecular weight is 103 kDa. Optimize starting dilution using relevant cell lysates (e.g., Jurkat, HeLa) from human, mouse, or rat origin.
- The antibody is listed as KO validated; consider using ITCH knockout cell lines to verify specificity in your experimental system.
- For ELISA, validate coating concentrations and detection antibody conditions with recombinant ITCH protein or known positive samples.
