Biological Background: ISCU Function and Localization
- ISCU is an iron-sulfur cluster assembly enzyme (also known as NifU-like protein) that serves as a scaffold for the de novo synthesis of [2Fe-2S] clusters in mitochondria. Related references: PMID:34824239, PMID:24971490, PMID:29576242, PMID:30031876
- ISCU exists in two interconverting conformational states—structured (S) and disordered (D)—which may regulate its function in cluster assembly. Related references: PMID:23940031
- It is a component of the mitochondrial core ISC assembly complex, where persulfide from the cysteine desulfurase NFS1 is transferred onto ISCU in an FXN-dependent manner, followed by [2Fe-2S] cluster formation with FDX2. Related references: PMID:24971490, PMID:29576242, PMID:30031876
- ISCU modulates NFS1 desulfurase activity in a zinc-dependent fashion and facilitates the interaction between frataxin (FXN) and the cysteine desulfurase complex. Related references: PMID:30031876, PMID:29576242
- A cytoplasmic isoform of ISCU functions in the extramitochondrial iron-sulfur protein biogenesis pathway, indicating dual mitochondrial and cytoplasmic roles.
- The gene is alternatively spliced, yielding isoforms that localize to mitochondria, cytoplasm, and nucleus, with the mitochondrial form containing a transit peptide.
- ISCU is widely expressed, with detection in heart, liver, skeletal muscle, brain, pancreas, kidney, lung, and placenta.
- Post-translational modifications include phosphorylation, and the protein binds iron as a metal cofactor essential for cluster assembly.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–167), which includes the mitochondrial transit peptide; consider this for isoform specificity in your assay.
- For WB, the expected molecular weight is approximately 18 kDa; validate with appropriate controls in human cell lysates.
- ELISA applications may require optimization of coating antigen and detection conditions; consider using recombinant ISCU as a standard.
- Since ISCU has both mitochondrial and cytoplasmic isoforms, subcellular fractionation may help reduce background in complex samples.
