Biological Background: IRS1 Function and Localization
- IRS1 (Insulin receptor substrate 1) is a signaling adapter protein that mediates signal transduction from the insulin receptor (INSR) and insulin-like growth factor I receptor (IGF1R). Related references: PMID:7541045, PMID:33991522, PMID:38625937
- Plays an essential role in development, growth, glucose homeostasis, and lipid metabolism. Related references: PMID:19639489
- Upon phosphorylation by the insulin receptor, IRS1 serves as a scaffold, engaging SH2 domain-containing proteins such as the p85 regulatory subunit of PI3K, NCK1, NCK2, GRB2, and SHP2. Related references: PMID:11171109, PMID:8265614
- Recruitment of GRB2 activates the Ras/Raf/MEK/MAPK cascade, while PI3K binding triggers the AKT pathway, collectively regulating cell growth, differentiation, and metabolic responses.
- Acts as a positive regulator of the Wnt/β-catenin signaling pathway by inhibiting DVL2 autophagy, thereby promoting cell proliferation. Related references: PMID:24616100
- IRS1 is present in both the cytoplasm and nucleus.
- IRS1 is a phosphoprotein implicated in diabetes mellitus; genetic variants are associated with disease susceptibility.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 1043–1242), which contains critical tyrosine phosphorylation sites for downstream signaling; consider this when designing phospho-specific or interaction studies.
- For Western blot, IRS1 migrates at approximately 132 kDa; use an appropriate gel percentage and optimize transfer conditions for high-molecular-weight proteins.
- In IHC-P, antigen retrieval may be necessary; validate in tissues known to express IRS1, such as liver, skeletal muscle, or adipose tissue.
- For IF/ICC, permeabilization conditions should be tested to detect both cytoplasmic and nuclear pools.
- The antibody cross-reacts with mouse and rat IRS1, enabling multispecies studies; verify sequence homology in the epitope region if extending to other species.
