Biological Background: IRP2 Function and Localization
- IRP2 (iron-responsive element-binding protein 2), also known as iron regulatory protein 2, is encoded by the IREB2 gene. It plays a critical role in cellular iron homeostasis.
- It binds specifically to iron-responsive elements (IREs) in the untranslated regions (UTRs) of target mRNAs, such as ferritin and transferrin receptor, to post-transcriptionally regulate iron uptake, storage, and utilization.
- This RNA-binding protein represses the translation of ferritin mRNA when iron levels are low, while simultaneously stabilizing transferrin receptor mRNA to increase iron import.
- IRP2 contains a 4Fe-4S iron-sulfur cluster and belongs to the aconitase family, consistent with its metal-binding and catalytic properties.
- The protein predominantly localizes to the cytoplasm, where it senses intracellular iron levels and modulates gene expression accordingly.
- Alternative splicing generates multiple isoforms, and post-translational modifications, including ubiquitination (Ubl conjugation), regulate its stability and activity.

- This antibody has been validated for Western blot (WB), immunohistochemistry on paraffin-embedded sections (IHC-P), and ELISA. In WB, a specific band around 105 kDa is expected. Due to preserved cross-reactivity with mouse and rat, it is suitable for comparative studies across these species.
- Optimal working concentrations should be determined empirically. Include appropriate positive and negative controls, especially when exploring tissue-specific expression or disease models.
