Biological Background: IRF6 Function and Localization
- IRF6 (Interferon regulatory factor 6) is a member of the interferon regulatory transcription factor family, and this antibody recognizes human IRF6 (UniProt: O14896). It also detects mouse and rat IRF6.
- IRF6 acts as a probable DNA-binding transcriptional activator. It is a key determinant of the keratinocyte proliferation-differentiation switch, playing a critical role in appropriate epidermal development.
- IRF6 may also regulate the proliferation of mammary epithelial cells, and it has been implicated in transcriptional control of WDR65.
- The protein is localized in both the nucleus and cytoplasm, consistent with its role as a transcription factor that may shuttle between compartments.
- IRF6 is expressed in normal mammary epithelial cells, but expression is significantly reduced or absent in breast carcinomas, suggesting a tumor-suppressive role.
- Post-translational modifications include ubiquitination (Ubl conjugation), and alternative splicing produces multiple transcript variants.
- The immunogen corresponds to amino acids 115-300, which covers the central region of the IRF6 protein, including part of the DNA-binding domain and flanking sequences.
Experimental Guidance and Technical Tips
- The antibody is validated for Western blot (WB) and ELISA. For WB, consider using whole-cell lysates from epidermal or mammary epithelial cell lines (e.g., HaCaT, MCF-10A) after confirming endogenous IRF6 expression.
- The immunogen targets the central region (aa 115-300) of human IRF6, which is highly conserved across human, mouse, and rat, supporting its cross-reactivity. Verify band specificity at the expected molecular weight of approximately 53 kDa.
- For ELISA, the antibody can be used as a capture or detection reagent; however, optimal working concentrations should be determined empirically in the specific assay system.
- Since IRF6 undergoes alternative splicing and may have multiple isoforms, a single band at ~53 kDa is expected for the full-length isoform in most samples, but additional bands may appear due to splice variants or post-translational modifications. Validate the pattern in your relevant model system.
